General description
Breast cancer anti-estrogen resistance protein 1 (UniProt P56945; also known as Cas scaffolding protein family member 1, Crk-associated substrate, p130Cas) is encoded by the BCAR1 (also known as CAS, CAS1, CASS1, CRKAS) gene (Gene ID 9564) in human. p130Cas/BCAR1 is an adaptor protein that belongs to the CAS family of scaffold proteins, members of which also include Nedd9, HEF-1 (CAs-L), EFS, and CASS4. Structurally, p130Cas is composed of a Src-homology 3 (SH3) domain, an adjacent large substrate-binding domain containing 15 repetitions of the YxxP motif, a main site of tyrosine phosphorylation on the p130Cas/BCAR1 molecule that generates SH2-binding sites, a proline and serine rich region, and a highly conserved four-helix bundle (focal adhesion targeting [FAT] domain). p130Cas/BCAR1 plays an important role in transducing signaling originated from mechanical stimuli, extracellular matrix (ECM) adhesion, as well as growth factor stimulation cascades during normal development and diseases.
